不同色泽甜樱桃果实花色苷积累与其相关酶活性之间的关系

摘 要:

为探明不同色泽甜樱桃果实发育期间花色苷的积累及其与相关酶活性之间的关系, 本研究以红色甜樱桃品种‘美早’和纯黄色品系‘13-33’为试材, 对果实发育期间花色苷总量及相关酶活性进行了测定, 并且采用超高效液相色谱-串联质谱技术(UPLC-MS/MS)对各花色苷组份进行了定性和定量分析。结果表明, 从‘美早’果实中检测出7种花色苷, 各组份含量伴随着果实的发育逐渐升高; 从‘13-33’中仅检测出4种花色苷, 各组份含量随果实的发育逐渐降低, 到果实成熟期其含量已甚微。两个品种的花色苷总量与各组份含量的变化趋势相同; ‘美早’中花色苷总量随着果实的发育逐渐升高, 特别是在转色期后急剧升高; ‘13-33’的花色苷总量随着果实的发育变化不明显, 并且一直维持在极低水平。‘美早’的PAL、CHI、DFR和UFGT酶活性在果实发育后期均显著高于‘13-33’。相关性分析结果表明, ‘美早’的花色苷总量与PAL和DFR酶活性之间无显著相关性, 而与CHI和UFGT酶活性呈显著正相关。

Relationship between anthocyanin accumulation and the activities of anthocyanin biosynthesis enzymes in different color of sweet cherry fruits

Abstract:

In order to explore the relationship between the anthocyanins accumulation and the activities of anthocyanins biosynthesis enzymes during development of the sweet cherry fruits, two sweet cherry cultivars Prunus avium ‘Tieton’ and ‘13-33’ with the different fruit color were used to detect the total anthocyanins content and anthocyanins biosynthesis related enzymes activities including plenylalanine ammonia lyase (PAL), chalcone ismoerase (CHI), dihydroflavonol 4-reductase (DFR) and flavonoid 3-O-glucosyltransferase (UFGT). The independent component analysis of anthocyanin was also detected using ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) in this study. The results showed that seven types of anthocyanins were identified in ‘Tieton’ fruits and the content of each component increased during the fruit development period, while four types of anthocyanins were identified from ‘13-33’ fruits, the content of each component decreased during fruit development and anthocyanin was barely detectable in mature fruits. Comparison of total anthocyanins content in these two cultivars revealed similar changing trend during fruit maturity. The total contents of anthocyanins in ‘Tieton’ fruits increased during fruits development and approached to a very high level at the late stage of fruit maturity. However, low level of total anthocyanins contents were detected across ‘13-33’ fruits development period and no obvious changes was revealed. The activities of PAL, CHI, DFR and UFGT in ‘Tieton’ fruits were significantly higher than that in ‘13-33’ at the late stage of fruit development. Correlation analysis showed that no significant correlation was observed in anthocyanin accumulation with the changes of PAL and DFR activities in ‘Tieton’ fruits. However, it was significantly and positively correlated with the activities of CHI and UFGT.
